Follow up paper on SOS in vascular cells

• H2S and SOS are produced and extruded in smooth muscle cells 20 x that in endothelial cells

• Hypoxia increased the generation of sulfenic acids while decreasing hydrogen sulfide

Conclusions

Treatment of melanoma with Cu, Zn or Fe thiomaltol complexes causes a strong apoptotic response in melanoma, with Cu by far most active.

Apoptosis increases with superstoichiometric additions of Cu at the same Htma dosing, which correlates with an increase in intracellular Cu. 

Cu/Htma treatment causes rapid morphological changes in melanoma cells.

Cu/Htma was shown to rapidly accumulate in lysomes preceding apoptosis, using EM and confocal microscopy.

Use of fluorescent dithiomaltol Httma shows its accumulation in extracellular membrane and lysosomes.

Cu/Htma treatment inhibits proteasomal activityconcurrent with cell death.

Alumnus does good! 

Farmer lab alum Prof. Michael Cohen from U Oregon Health and Science gave a talk at UC Irvine and saw many familiar faces, including Prof. Ev Fleischer who used to be active in our lab.

Congrats again, Mike!

Michael Spiegel has defended and will graduate in December... his last paper was a long collaboration with the Solouki group on characterizing complex mixtures of asphaltenes, published in Energy and Fuels, 2018, 32 (10), pp 10549–10555
